I think your memory card should formatted FAT32. It sounds like a File System problem. Try to back up your stuff on your memory card and than format it using Windows and selecting FAT32 under the file system option. After that it should read/write properly on the PC. * +

windows should read it no problem, access denyed normaly means the antivirus has blocked some content on the phone. maybe an app or game. turn the av off and see if the problem persists. * +

if it doesnt it means theres a virus or malware on your phone. and it need scanning, syaing as you av has blocked it just scan with that... if that av is the cause. * +
